For this single-cell studies are essential, where the intrinsic behavior of cells can be examined. Current
single-cell technologies depend on genetic or phenotypical readouts, based on end-point analyses
only. In this study, a microfluidic platform is characterized, optimized, and applied, that allows for the
functional study of cytotoxicity of single cells in real time. Using droplet-based microfluidics, single
immune cells are co-encapsulated with tumor cells in picolitre-sized droplets to achieve complete
isolation of cells. This allows for the dynamic, high-throughput analysis of functional behavior at a
single-cell level. The platform’s potential is demonstrated in the study of cytotoxic activity of single
plasmacytoid dendritic cells (pDCs) against tumor cells. pDCs are a rare, heterogeneous immune cell
type with cytotoxic function and potential in immune therapy, which makes them a promising subject
